How it works

MK-677 (ibutamoren) is an orally active, nonpeptide growth hormone secretagogue that functions as a high-affinity agonist of the ghrelin receptor (GHS-R1a) in the hypothalamus and pituitary. Activation of GHS-R1a stimulates growth hormone (GH) release via phospholipase C–IP₃/Ca²⁺ signaling, increasing pulsatile GH secretion without suppressing endogenous rhythm. Elevated GH subsequently induces hepatic and peripheral production of insulin-like growth factor-1 (IGF-1), mediating downstream effects on protein synthesis, nitrogen retention, lipolysis, and bone metabolism, while largely bypassing direct androgen or estrogen receptor pathways.
