How it works

TAK-653 is a selective positive allosteric modulator (PAM) of AMPA-type ionotropic glutamate receptors (AMPARs) that potentiates endogenous glutamatergic neurotransmission without directly activating the receptor. By enhancing activity-dependent AMPAR signalling, TAK-653 facilitates long-term potentiation (LTP), upregulates BDNF–TrkB and downstream mTORC1 signalling pathways, and promotes synaptic plasticity, neurotrophic signalling, and functional neuronal connectivity associated with learning, memory, and mood regulation.
