Methylene Blue

Methylene blue is a redox-active compound investigated for its ability to enhance mitochondrial electron transport and reduce Reactive Oxygen Species. It may also modulate the Nitric Oxide–cGMP pathway and inhibit Monoamine oxidase, with potential, but not definitively established, cognitive-enhancing effects.

Methylene blue is a redox-active phenothiazine compound whose mechanism of action is multifactorial and not fully elucidated. It has been investigated as an alternative electron carrier within the Electron Transport Chain, where it may facilitate electron flux from NADH to downstream acceptors such as cytochrome c, potentially reducing electron leakage and modulating Reactive Oxygen Species generation.

It has also been proposed to inhibit the Nitric Oxide–cGMP pathway via suppression of soluble guanylate cyclase, resulting in decreased cGMP signaling. Additionally, it has been investigated as a reversible inhibitor of Monoamine oxidase, suggesting potential modulation of monoaminergic neurotransmission.

From a neurocognitive perspective, low-dose methylene blue has been investigated for possible enhancement of mitochondrial respiration and neuronal metabolic efficiency, particularly in high-energy-demand regions such as the hippocampus and prefrontal cortex. This may facilitate synaptic plasticity and memory consolidation processes, potentially via improved oxidative phosphorylation and reduced oxidative stress burden. Some experimental data suggest dose-dependent effects on attention, memory retention, and neural network efficiency; however, these findings remain preliminary and context-dependent, with higher concentrations potentially exerting opposing, pro-oxidant effects.
