How it works

Enclomiphene citrate is a nonsteroidal selective estrogen receptor modulator (SERM) that acts primarily as an estrogen receptor (ER) antagonist in the hypothalamus and pituitary. By competitively inhibiting estradiol-mediated negative feedback at ERα, it increases gonadotropin-releasing hormone (GnRH) pulsatility, leading to elevated secretion of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) from the anterior pituitary. This upregulation stimulates Leydig cell steroidogenesis and Sertoli cell function, resulting in increased endogenous testosterone production and spermatogenesis without direct androgen receptor activation.
